"Shakespearean" is the correct adjective to describe things associated with Shakespeare.

"Shakespearian" is technically a valid noun, which means a person associated with Shakespeare. However, it's much more common (and helpful) to use the adjective form along with a more specific noun, e.g. a Shakespearean actor, a Shakespearean scholar.

In both cases, the term is capitalised.

(Thanks to Duane for the information!)
